Genetic Diversity and Population Structure of Portunustrituberculatus in Released and Wild Populations Based on Microsatellite DNA Markers from the Yangtze Estuary

Portunus trituberculatus is an important economic species of crab that artificially bred and released in the Yangtze River Estuary its adjacent sea areas. Based on six microsatellite markers, we investigate genetic diversity structure 101 P. specimens collected from two hatcheries Nantong Zhoushan participated stock enhancement year 2019. We compared these with 124 wild caught 13 localities estuary. Analysis several parameters (NA, RS, I, HO, HE, FIS, FST) for 15 populations demonstrates both possess relatively rich diversity. Furthermore, groups demonstrate no less variation between themselves than do crabs. Most FIS values are greater zero, which shows inbreeding common among geographically open sites. However, insufficient sampling may have led to a wide distribution null alleles, Hardy–Weinberg test disequilibrium markers PN22 P04, lack site 14. All (except locality 14) not suffered bottleneck effect. Four subgroups can be seen roughly spread longitudinally along sample area by performing pairwise comparisons distance FST populations. No obvious topological heterogeneity discovered four phylogenetic tree. The existence exchange differentiation also verified using analysis.
